However, our previous work did include mCherry controls, and did not find an impact of CNO on punishment-related behav- iors (Li et al. 2025). Additionally, chemogenetic manipula- tions were tested using only one heroin dose. It is possible that alternative doses may have produced different behav- ioral outcomes. Furthermore, Saline/CNO injection order could not be fully counterbalanced; when attempted, CNO- induced behavioral alterations persisted into the following day, making full counterbalancing infeasible. However, because behavior was stable at the time of chemogenetic testing, we feel confident that the observed effects reflect CNO manipulations rather than effects of time. We acknowledge that subjects in the chemogenetic and electrophysiological experiments underwent different stress- induced reinstatement procedures, with only the chemoge- netic cohort exposed to the novel dual-hit paradigm. This difference reflects the fact that these studies were conducted in parallel as a part of separate experimental aims. It is pos- sible that exposure to an additional stressor could further alter DRN 5-HT electrophysiological profiles, potentially yielding different outcomes under dual-stress conditions. Moreover, an outstanding question concerns the temporal emergence of these electrophysiological alterations. Previ- ous work in our laboratory examining stress-induced rein- statement of cocaine seeking and morphine conditioned place-preference also revealed enhanced GABAergic sig- naling within the DRN. These changes only emerged with combined drug and stress history, and were not present with 1 3Psychopharmacology either factor alone (Staub et al. 2012; Li and Kirby 2016); we believe that a similar interaction may be driving effects observed here. However, expanding recording time points would facilitate the identification of specific experiential fac- tors, such as drug or stress exposure, that may differentially shape DRN electrophysiological adaptations over time. We examined only activation of the DRN 5-HT system and did not assess behavioral consequences of Gi-mediated inhibition of DRN 5-HT neurons. Across prior studies in our laboratory, inhibition of the DRN 5-HT system has not produced behavioral effects (Li et al. 2025; McElroy et al. 2025; McCloskey et al. 2025), which we believe may be attributable to low basal 5-HT levels in the brain and a functional “floor” that limits the impact of further inhibition (Rueter et al. 1997; Gonzalez et al. 2003; Tao and Ma 2012). Based on this prior experience and the consistent lack of behavioral effects following inhibitory manipulations in our hands, we focused our resources on Gq-mediated activation in the present work.
