  • Thymus vulgaris and Thymus serpyllum: extraction, distillation, and encapsulation strategies, phytochemical composition and bioactivity (2026) · Phytochemistry Reviews · doi

This review provides a comprehensive and compara- tive overview of T. vulgaris and T. serpyllum essential oils, extracts, and major bioactive compounds, focus- ing on extraction and distillation strategies, phyto- chemical composition, biological activities, and encapsulation approaches. Both species are character- ized by the presence of monoterpenes (e.g., thymol, carvacrol, linalool, and c-terpinene) and polyphenolic compounds (e.g., rosmarinic acid and flavonoids), whose concentrations vary depending on chemotype, geographical origin, environmental conditions, har- vesting stage, and extraction methodology. These factors represent the primary sources of variability in reported phytochemical profiles and limit a direct comparison between studies. Reported biological activities, including antioxi- dant, antimicrobial, anti-inflammatory, cytotoxic, and anticarcinogenic effects, were predominantly investi- gated using in in vitro assays and preclinical models. However, differences in experimental design, sample 123 preparation, assay conditions, and reporting units limit cross-study comparability and definitive conclusions regarding the relative bioactivity of T. vulgaris and T. serpyllum extracts and essential oils. Moreover, the observed biological effects arise from complex inter- actions within essential oils and extracts and should not be directly inferred from studies on isolated compounds alone due to matrix interactions and differences in concentration and bioavailability. Extraction methods remain a critical determinant of both the composition and functionality of T. vulgaris and T. serpyllum extracts and essential oils. While conventional hydrodistillation and steam distillation are widely used, emerging green extraction tech- niques, such as supercritical fluid extraction and ultrasound-assisted methods, offer improved effi- ciency and selectivity. However, further work is required to standardize extraction protocols and eval- uate their reproducibility across different plant mate- rials and conditions. Similarly, although encapsulation strategies, such as polymeric and lipid-based systems, show potential for improving stability and delivery, their effectiveness is highly dependent on formulation design and remains insufficiently validated in complex biological systems. Finally, the influence of synergis- tic and antagonistic interactions within complex mixtures requires further clarification using integra- tive experimental and computational approaches. Future research should therefore prioritize (1) standardization of extraction and analytical protocols, (2) harmonization of bioactivity assays and reporting units, (3) mechanistic studies combining experimental and computational methods, and (4) validation of promising findings in relevant in vivo models and safety assessments. In this context, computational approaches such as molecular docking, molecular dynamics, and quantum–mechanical calculations may into interaction mecha- provide additional nisms, complementing the experimental validation (Furlan et al. 2018, 2024; 2026; Furlan and Bren 2021; Kores et al. 2022). insight In conclusion, both T. vulgaris and T. serpyllum essential oils and extracts demonstrated significant health-beneficial biological activities in preclinical experimental systems due to the presence of various bioactive compounds. However, the observed differ- ences in phytochemical and bioactivity profiles are not strictly species-dependent, but are strongly modulated by chemotype, geographical origin, environmental
